The 47-year-old is to step down at the end of the season, having led Dortmund to the Bundesliga title in 2011 and a league and cup double in 2012.
"To crown this amazing time with a such a moment would only be fair," said Klopp, who was appointed in 2008.
Wolfsburg, who finished second in the league, 10 points behind winners Bayern Munich, have never won the German Cup.
The Wolves, whose only league title came in 2009, reached the final in 1995 but lost 3-0 to Borussia Monchengladbach.
Dieter Hecking's side will pay tribute to former player Junior Malanda,who was killed in a car accident aged 20 in January, by wearing a special shirt for the match in Berlin.
Dortmund, Champions League runners-up in 2013, were bottom of the German league table in February but recovered to secure seventh and a Europa League place with victory over Werder Bremen in Klopp'sfinal home game in charge.
"It was our target at the start of the season to be here now and I'm happy we are after such a difficult season," added Klopp.
"We still have something left to do. It's not my aim to be the hero tomorrow - I'm happy to leave that to the players."
The match will also be Sebastian Kehl's last for Dortmund, with the 35-year-old midfielder set to retire at the end of the season after 14 years with the club.
